Gregg L. Semenza, M.D., Ph.D.

Dr. Semenza has identified and characterized hypoxia-inducible factor 1 (HIF-1), a basic-helix-loop-helix transcription factor. HIF-1 expression increases exponentially as cellular O2 concentration declines. HIF-1 expression and activity are regulated by hydroxylaton of proline and asparagine residues, a novel O2-dependent post-translational modification.

 

HIF-1 activates transcription of genes that are essential for adaptive responses to hypoxia. Multiple projects are ongoing in the laboratory to investigate the role of HIF-1 in development, physiology, and disease pathophysiology.

These investigations involve analyses of tissue culture cells, mouse models, and human pathological specimens. HIF-1 is overexpressed in many common cancers and its overexpression is associated with tumor angiogenesis, invasion, metastasis, radiation resistance, and patient mortality. He is exploring the mechanisms and consequences of HIF-1 overexpression in human cancer. His goals are to develop novel diagnostic, prognostic, and therapeutic strategies that target HIF-1.
